The generation of double-stranded cDNA is a crucial step during NGS library preparation from RNA. Tailing and ligation of cDNA -TLC- presents a novel approach to generate full-length cDNA molecules, offering an alternative to template switch oligos.
This work will showcase the potential of TLC through proof-of-concept experiments for additional applications, including full-length cDNA sequencing, profiling of RNA modifications, and its applicability to low input samples and single cells.
